    R-Phycoerythrin (R-PE) was originally isolated from red algae and has thress types of subunits: α (~ 20,000 daltons), β (~ 20,000 daltons) and γ (~ 30,000 daltons). The molecular weight of intact R-PE has been found to be about 240,000 daltons, and a subunit structure of (αβ)6γ has been determined. Its primary absorbance peak occurs at 565nm with secondary peaks at 495 and 545nm; the relative prominence of the secondary peaks varies significantly among R-PEs from different species. RPE has been used in a variety of immunological assays and as a fluorescent label for Fluorescence Activated Cell Sorting (FACS). Nonetheless, by virtue of its high absorption coefficient and almost perfect quantum efficiency, it is one of the brightest dyes available today. In addition, because of the high molar absorptivity of these proteins at visible wavelengths, they are convenient markers in such applications as gel electrophoresis, isoelectric focusing and gel exclusion chromatography. Sulfo-SMCC are heterobifunctional crosslinkers that contain N-hydroxysuccinimide (NHS) ester and maleimide groups. NHS esters react with primary amines of R-PE at pH 7-9 to form amide bonds, while maleimides react with sulfhydryl groups at pH 6.5-7.5 to form stable thioether bonds. SMCC-activated R-PE is a stable protein which can be easily linked to antibodies and other sulfhydryl-containing proteins without altering its spectral characteristics. Excess non-reacted Sulfo-SMCC and by-products have been removed.

    Allophycocyanin (APC) is a phycobiliprotein isolated from Spirulina sp., a blue-green alga. APC has α, β (~ 17,000 daltons) two types of subunits. The molecular weight of intact APC has been found to be about 105,000 daltons. Allophycocyanin is the least stable among the major phycobiliproteins, susceptible to dissociation at low concentrations including concentrations at which some assays are performed. CL-APC is chemically cross-linked between α and β subunits, and is much more stable than APC. The crosslinked allophycocyanin has improved stability in aqueous solution. CL-APC primary absorbance peak occurs at 650nm, and can be excited by either a dye laser (595-605nm), or the less expensive HeNe laser (633nm). Cross-linked APC is emerging as one of the preferred fluorescent probes for Fluorescence Activated Cell Sorting (FACS) and Forster-type Fluorescence Resonance Energy Transfer (FRET) assays. Nonetheless, by virtue of its high absorption coefficient and almost perfect quantum efficiency and emission in a region of the spectrum with extremely low cellular autofluorescence, it is one of the brightest dyes available today.
