第四军医大学樊代明院士课题组在《Theranostics》发表了题为“O-GlcNAcylation of SIX1 enhances its stability and promotes Hepatocellular Carcinoma Proliferation”的文章。代谢异常是肿瘤生长中的标志之一。然而，癌细胞是如何协调葡萄糖代谢和肿瘤的增殖在很大程度上未知的。本项研究中，作者证明了调节Warburg效应和癌症的关键转录因子SIX1在体内外促进了肝癌细胞的生长。除了调节糖酵解通路外，作者还发现SIX1可以提高O-GlcNAc糖基化水平。重要的是，SIX1本身也被O-GlcNAc糖基化修饰，并且糖基化修饰抑制了SIX1的泛素化降解。此外，位点定向诱变T276A可降低O-GlcNAc糖基化水平，逆转SIX1的促肿瘤作用。本项研究说明O-GlcNAc糖基化 SIX1是协调葡萄糖代谢和肿瘤进展的重要方式。
