暨南大学刘杰教授课题组在《Applied Materials Today》发表了题为Porous selenium nanozymes targeted scavenging ROS synchronize therapy local inflammation and sepsis injury的文章。

 

目前脓毒症发病率和死亡率逐年上升,其主要因素是炎性巨噬细胞内产生过量的活性氧(ROS)会导致局部炎症损伤和脓毒症型器官功能障碍,而人体内缺乏有效的ROS清除剂。因此,开发高效的ROS清除剂是治疗局部炎性和脓毒症损伤的有效策略。

 

在此,该课题组构建了具有模拟谷胱甘肽过氧化物酶(GPx)活性的介孔硒纳米酶治疗体系(MSe-HA NPs),通过靶向清除炎性巨噬细胞内的ROS治疗局部炎症和脓毒症双重损伤。这种具有较大比表面积(1160.195 m2 /g)MSe NPs相比于实心Se NPs,提供了更多活性位点,催化反应速度提高了2.02倍,有效提升了H2O2清除效率。

 

此外, MSe NPs还能够通过清除具有损伤作用的超氧阴离子(×O2-)和羟基自由基(×OH) 表现出广谱的ROS消除能力。利用透明质酸(HA)修饰获得MSe-HA NPs,通过特异性结合CD44受体增强了MSe NPs对炎性巨噬细胞的靶向性,此外,HAROS降解特性有效保证了MSe NPsROS接触后,重新暴露介孔结构并发挥催化作用清除细胞内的ROS,保护正常细胞免受炎症损害。在体内治疗中,进一步证实了MSe-HA NPs可以通过迅速消除炎症区域内过量的ROS,有效治疗局部炎症和脓毒症的多种损伤。

 

因此,这种新型介孔纳米酶为纳米药物的设计和治疗炎症相关疾病提供了有效策略。
